Interesting, you do the fills first? I always do the outlines first in vector pieces, then the fills and hilights and shadows last.

Actual dpi is not that important as the pixel dimensions but a sufficient resolution for a shirt print would be 300dpi at the intended physical size. So a 12 inch wide image would be 3600px wide. It can always be bigger too and that might be a good idea if you want to use the image for bigger prints also.
